The PHP SCB core is equipped with a NanoCellect® WOLF G2. Cell Sorter. This instrument enables MSU researchers to gently sort engineered and primary cell lines, stem cells/iPSC, dissociated cells from organoids or tumor tissues for healthier intact post-sort growth and downstream analyses. Aerosol-free microfluidic cartridge and tissue-culture hood encasing of compact instrument enables contaminant- and biohazard-free environment to dispense single cells from plant, animal and human sources.
The WOLF G2 Blue 488 nm/Yellow 561 nm laser configuration affords < 250 MESF sensitivity and co-detection of up to 7 fluorochrome tagged markers and/or fluorescent proteins (eGFP, tdTOMATO, RFP) in a single cell. This innovative platform enables detailed profiling and parallel gentle sorting of up to two cell subpopulations with translational implications for personalized and precision medicine.
Versatile configuration of the instruments enables single cell dispensing in a 96-well format, bulk soring in 5 mL tube (up to 1 million cells) and 10,000+ event cytometric analysis. Application include CRISPR gene editing, cell line development, preparation for single-cell DNAseq and RNAseq, Immunology and Antibody Discovery, single-cell cancer cell heterogeneity and analysis, gentle sorting of hepatocytes, fibroblasts, neurons, plant cells.
